Caracterização morfológica, reprodutiva e genômica em indivíduos de Psidium cattleyanum Sabine (Myrtaceae) com diferentes conteúdos de DNA

Psidium cattleyanum Sabine, popularly known as araçá da praia or araçá de crown, is a native species present mainly on the Brazilian coast and currently cultivated in several countries. Studies on the species are recent and have been gaining prominence mainly in the pharmaceutical and food sector. The species draws attention for having different chromosome numbers, such as 2n = 44, 55, 66, 77, 88, 99, 110 and 132, described in the scientific literature. These differences in ploidy may imply morphological, anatomical and molecular variations as already reported in other angiosperms. The objective of this work was to carry out the morphological and micromorphological characterization of the leaves, to analyze the genomic data and to evaluate the pollen viability, in comparison to the different DNA contents present in the individuals of P. cattleyanum. For that, the external morphology and micromorphology of leaves of nine individuals that are part of the germplasm collection of CCAE/UFES, Campus de Alegre, Espírito Santo, Brazil, were analyzed. The study of external morphology comprised the analysis of dry mass, length and width of the leaf blade and petiole of nine leaves per individual. For micromorphological analysis, the leaves were collected and fixed in FAA 70% for 24 h and stored in 70% ethyl alcohol, then diaphanized and stained with safranin. Five slides from the median region of the abaxial face were prepared for each individual to perform the stomatal density count and stomata size. The genomic data used to perform the identification of polymorphism between individuals were taken from the database of the Biometrics Laboratory of PPGGM - UFES. And for pollen viability analysis, flower buds were collected in the pre-anthesis phase, stored in the freezer with methanol and acetic acid (3:1). To make the slides, a cross-section of the floral bud was performed, followed by staining with Alexander dye and counting one thousand pollen grains per slide in a total of three slides per individual. The data obtained were tabulated in an electronic spreadsheet for the construction of descriptions and performance of multivariate analyses. The differences observed in the analyzed characteristics between individuals with higher DNA contents (7.03 and 6.83 pg), lower DNA contents (3.31 and 3.45 pg) and median DNA content (3.66, 3.71, 3.77, 4.26 and 4.71 pg), demonstrated that the relationship between individuals with higher DNA content presenting morphological and micromorphological characteristics of larger leaves was not identified. In the pollen viability analysis, individuals with DNA content ranging between 3.8 and 3.9 picograms of DNA had a low number of viable pollen. And in the analysis of genomic data, individuals with intermediate DNA content showed the greatest variations.
