Resumo
The cellulases are enzymes used in several segments of the industry, and among its applications the production of biofuels using the enzymatic hydrolysis of lignocellulosic materials, from agroindustrial residues (cocoa, coffee, orange, and others peels) is noteworthy. However, the major obstacle to the expansion and commercialization of this production corresponds to the high cost of cellulases production. In this context, the aim of this work was to study the solid state fermentation conditions using the filamentous fungus Trichoderma harzianum for cellulases production and applying cocoa biomass as substrate. Cultures in erlenmeyer flasks were carried out to evaluate the influence of moisture content, in the range of 50 to 70%, and of the proportion of cocoa peel (CP), in the range of 40 to 60%, applying an experimental design 32 , being the enzymatic activity response variable. The influence of aeration, using wheat bran and sugarcane bagasse, on enzyme production was evaluated by measuring the FPase enzymatic activity in Raimbault columns with forced aeration, comparing with static cultures under the same conditions. A maximum FPase enzyme activity of 1.85 U/gds was obtained with 60% CP and 70% of moisture in 120 hours of culture in flasks, for which moisture was the factor that caused the greatest effect. In the column fermentations, FPase activity reached 5.40 U/gds against 3.50 U/gds in the static fermentation, corresponding to an increase of 51.3%. Therefore, CC proved to be promising as a substrate for FES cellulase production and forced aeration made it possible to obtain larger enzymatic activities, thus confirming its strong influence on FES processes.
