Resumo
Tannase is an industrially important enzyme and has several applications in the food, animal feed, beverage, pharmaceutical and chemical industries. It can be obtained from animal, vegetable and microbial sources, being the microbiological medium main source of this enzyme obtaining. Among microorganisms, fungi stand out in the tannase production due to their versatility in hydrolysis of tannins different types. Objectives of this work were selection among fungi Phanerochaete chrysosporium, Aspergillus niger, Penicillium sp. and Fusarium oxysporum to determine the best tannase producer; enzyme production by solid state fermentation (SSF) with use of eucalyptus bark (EB) and wheat bran (WB) as substrates; and statistical analysis for production optimization. Penicillium sp. was selected for the tannase production tests under the different cultivation conditions in SSF. SSF conditions optimization was carried out through a Central Composite Rotational Design (CCRD), from study the influence of moisture content, EB and WB proportion and tannic acid concentration on the tannase enzymatic activity. According to regression model generated in CCRD, the maximum tannase enzymatic activity was 26.57 U.gms-1 , under the condition 80% moisture on a wet basis, 50% EB and 50% WB, and tannic acid concentration of 25.0 g.L-1 . Therefore, eucalypt bark can be transformed into a product with high added value, since it was a promising substrate for the tannases production by SSF.
